Factors Influencing the Components of the Total Protonmotive Force in Mung Bean Mitochondria

Abstract
The components of the protonmotive force (Δp) have been determined for mung bean mitochondria under various metabolic states using an ion distribution technique. Under state 4 conditions, a Δp of 180 mV is attained which is reduced to 150 mV during state 3 respiration. The addition of Ca2+ during state 4 conditions had no effect on either component of Δp, whilst an uncoupling agent such as 1799 completely collapsed both components. The hydrolysis of exogenous ATP generated a potential which was consistently 50 mV lower than that obtained under state 4 conditions. In the presence of 100 mM KCl, the value of Δp was 79 mV, the values of -ZΔpH and ΔΨ being 48 and 31 mV respectively.
