Inactivation of Enzymes by Visible Light in the Presence of Riboflavin

Abstract
Investigations were conducted to determine whether riboflavin sensitized the photoinactivation of alpha-amalyse. The diluted enzyme was mixed with phosphate buffer lacking or containing 100 [mu]g. %, riboflavin (Rbf), stored in the dark or illuminated by 400 ft.-candles of fluorescent light for 1 hr., and then tested appropirately for inactivation. Visible light or Rbf applied separately had slight effect on rate of enzyme action, but illumination of enzyme solns. containing Rbf resulted in marked reduction of enzyme activity. Studies on the kinetics of the inactivation reaction indicated that it appeared to be a first-order reaction.
