CONSTRUCTION OF FUNCTIONAL RECOMBINANTS (HYBRID) OF DNA-MOLECULES INVITRO

Abstract
The preparation of a hybrid plasmid containing DNA from the ColE1 plasmid and the R6K plasmid is described. EcoRI restriction endonuclease from Escherichia coli KM182 and DNA ligase from phage T4 am81 infected E. coli B were used. The hybrid plasmid was introduced by transformation into E. coli C6000. Analysis of 1 of the transformed clones indicated that the recombinant plasmid contained ColE1 integrated within the R6K region that determines transmissibility.
