A Surface Plating Technic for Determining Bacterial Population of Milk

Abstract
A drop plate technique for determining total counts of milk is present. Tryptose glucose extract agar plates are dried, inverted with bottoms raised slightly for 4 hours at 35[degree]C. Dilutions of the milk are made in buffered water containing 1-20.000 Triton x 100. One-tenth ml portions are delivered to quadrants of the agar plates with a 0.2 ml pipette. After the drops on the plates have been absorbed in the agar, the plates are incubated at 35[degree] C for 36-48 hours in an inverted position. The bacterial counts are comparable to those obtained by pour plates using the same medium.Keywords
