Demonstration of Myosin in Human Striated Muscle by Fluorescent Antibody

Abstract
Anti-myosin sera were obtained by injecting rabbits with purified human myosin. The gamma-globulin fraction of the sera containing high antibody titers was labeled with fluorescein isocyanate according to the Coons'' technique. Human, cat, rat, mouse and rabbit muscle were stained with fluorescent antihuman myosin antibodies. Observations were made on tested single fibers or on muscle tissue fixed briefly in formalin, embedded in diethylene glycol disterate and sectioned at 6 u. Also, some tissues were instantaneously frozen in isopentane and sectioned in the cryostat. Striated muscle fibers exhibited bright fluorescence of A bands. In the heart muscle, the intercalated discs remained non-fluorescent. In human muscle undergoing degeneration, bright fluorescence was observed in atrophied diseased fibers.Keywords
