Gas-Liquid Chromatographic Determination of Rotenone and Deguelin in Extracts of Tephrosia vogelii

Abstract
Treatment of rotenone with sodium borohydride gives a derivative that produces a sharp elution peak under gas-liquid chromatographic (GLC) conditions that lead to broad multiple peaks from rotenone itself. The reaction can be carried out rapidly on a small scale and the product injected directly onto the column after a simple workup. When used with a mixture of rotenone and deguelin, such a reduction-GLC sequence provides two distinct peaks. Applicability of the procedure to determine these compounds in crude Tephrosia vogelii extracts is demonstrated. Qualitative results for other rotenoids are also reported.
