Study of Respiratory Chain in Small Amounts of Biological Materials

Abstract
The possibility of applying the spectrophotometric method for assay of mitochondrial oxygen uptake, based on the utilization of oxyhemoglobin as the oxygen donor and indicator, in human biochemistry is studied. The special problems posed by such a study are the following: (1) achieving of sufficiently large sensitivity for accurate measurements of the oxygen uptake by about 1 mg of tissue ; (2) the knowledge of enzymatic properties of mitochondria in diluted suspensions as preventing their dénaturation; (3) to find a suitable method for ATP synthesis evaluation (P/0 ratio), and (4) the adaptation of the spectrophotometric method, as well as the oxygen cathode method, to dynamic studies of tissular respiration.
