A Thermo-gradient Apparatus Designed for Biological Investigations over Controlled Temperature Ranges

Abstract
A description is provided of apparatus designed to produce continuous temperature gradients over selected variable ranges between 0 and 50 °C. The unit was constructed for seed-germination studies, but would be suitable for a wide range of physiological, mycological, or biochemical investigations. The basic unit is an aluminium bar insulated on three sides with expanded polystyrene, heated electrically at one end and cooled at the other with a semi-conductor heat pump. The temperature range from one end of the bar to the other is determined by the balance between heat input and removal, controlled separately at the hot and cold ends of the bar. Methods are described for maintaining stabilized temperatures along the bar for long periods, and for the regular monitoring of temperatures in a unit of six thermo-gradient bars.Keywords
