Blastocyst Expansion and Hatching of Bovine Ova Cultured in Vitro

Abstract
Sixty-three bovine embryos between the one- and eight-cell stage were collected from synchronized, PMSG treated heifers. Embryos were placed randomly in culture among two media supplemented with heat treated fetal calf (HTFC) serum or bovine serum albumin (BSA) and development was recorded at 24-hr intervals for the duration of culture (130 to 160 hr). Blastocyst expansion and hatching was achieved by two- and eight-cell bovine embryos when cultured in Modified Ham's F-10 or Ham's F-10 medium supplemented with 10% HTFC serum. No development past early blastocyst was achieved by one-, two- or eight-cell embryos when 1.5% BSA was used as a supplement to modified Ham's F-10 or Ham's F-10 medium. The results suggest that factors present in HTFC serum appear to be required by bovine embryos for blastocyst expansion and hatching in vitro. Copyright © 1976. American Society of Animal Science .
