Abstract
The technique allows one to obtain orientated microtome sections from a large or small sample as well as a tissue strip in the area adjacent to the microtome section from the same sample. The strip can be handled like a whole mount, and the different cell layers can be viewed by differential interference-contrast (Nomarski) microscopy. The strip in combination with the available microtome sections gives a total picture of the tissues in question. The technique offers great advantages in many developmental or purely histological studies, especially in the study of complicated dorsiventral flowers, as demonstrated by our example.
