Abstract
Ro 3-3528 (racemic 6,7-dimethyl-[alpha]-[(isopropylamino)methyl]-2-benzofuranmeth-anol hydrochloride) had 3 primary pharmacological actions: A strong and selective [beta] -adrenolytic action is demonstrated by a competitive antagonism of the positive chronotropic and positive inotropic effects of catecholamines in isolated atria and the heart in situ and by the inhibition of the hypotensive effect of isoproterenol; An antifibrillatory action is shown by a lengthening of the effective refractory period of isolated guinea-pig atria and the reduction of cardiac glycoside toxicity on the heart in situ and in vitro. A local anesthetic potency 2.5 times greater than that of procaine was found in studies in isolated rabbit saphenus nerves.Keywords
