Abstract
Clean healthy roots are obtained by excising a few tillers very close to their point of emergence from the base of a larger plant and then growing them in vermiculite to which is periodically added a quantity of a complete mineral nutrient solution, sufficient to keep the medium moist. The roots are treated, while still intact on the living plant, with 0.0015 M 8-hydroxyquinoline for 4 hours at 18°C. At the end of treatment, the roots are excised and placed in 45% acetic acid for approximately 5 minutes, after which the root-tips are stained in Feulgen. Thereafter, temporary squash preparations are made of the meristems.Keywords
