Preparation of Cytochrome c with the Aid of Ion Exchange Resin.

Abstract
Cytochrome c (I) was prepd. according to Keilin and Hartree (Biochem. Jour. 39: 289. 1945.) and chromatographed on conditioned cation exchanger (Amberlite IRC - 50). I lodged at the top of the column and on every trial, a golden-colored impurity, protein in nature, appeared quickly in the effluent. I was eluted 0.1 [image] NH4OH - NH4 acetate at pH 10.8. The original cytochrome c contained 0.24% Fe, and the front and tail fractions from the chromatogram contained 0.447% and 0.461% Fe. Other prepns. always separated into several fractions, each of which contain higher levels of Fe. The main component is a very highly purified substance as verified by light absorption values and electrophoretic migration.This publication has 2 references indexed in Scilit:
