A Method of Conducting the 50 Percent Hemolysis End Point Complement-Fixation Test for Parasitic Diseases

Abstract
In a sensitive method of conducting complement-fixation tests on helminth and protozoan diseases, four 50% units of complement are used. The unit of complement is detd. by comparing the tubes of the complement titration with the 50% hemolysis tube of a prepd. color standard. The diagnostic test is conducted in a total volume of 1 ml., the mixture of serum, complement and antigen dilutions in 0.20 ml. amts., respectively, being refrigerated overnight. The 0.4 ml. of sensitized cell are added the following morning and the tests are set in a water bath for 30 min. at 37 [degree]C. For each serum tested a quantitative complement-deterioration test is employed. By this means the amt. of complement which deteriorates in the refrigerated tests in the presence of each serum dilution alone can be readily detd. This deteriorating factor is called the "binding power" of that particular serum dilution and must be considered in evaluating the test. The same check is made on each antigen dilution used and the binding power of the antigen dilutions must also be considered in the final evaluation.Keywords
