Determination of small amounts of phosphatides and cholesterol in tissues

Abstract
Methods are described for the extraction and estimation of phosphatides and cholesterol in 5-20 gm. of tissue. Chloroform does not extract lipid substances completely from dried tissues. MgCl2 precipitates pure lecithin almost completely from solution in light petroleum and acetone. A high fat content interferes with the precipitation by MgCl2. The micro-gravimetric method for pure cholesterol was applied to tissue extracts. Fat interfered with the free cholesterol determinations but did not seem to interfere with total cholesterol if dissolved in ether.
