Abstract
A study was made of the antibacterial and pharmacological activity of oxolinic acid (Prodoxol), a new synthetic antimicrobial agent used in the treatment of urinary tract infections. 95% of the strains of Eseherichia coli, Proteus mirabilis and indol-epositive Proteus studied were sensitive to 031 μg/ml. Strains of Kiebsiella/Enterobacter were less sensitive, only 61% being inhibited by 031 μg/ml. Strains of Pseudomonas aeruginosa and Streptococcus faecoiis were highly resistant. The mean peak concentration in the serum of volunteers on an oral dose of 750 mg twice daily was 08 μg/ml; in the urine, it was 197 μg/ml with 1% of each dose excreted in an active form over 8 h. These levels were low, but inhibitory to the most common urinary pathogens. The infrequent dosage is clinically convenient, but the incidence of side effects in volunteers was high. Cost and toxicity dictate that it should not be used as a first-line drug in the therapy of urinary tract infections.Keywords
