Genotypic Differences in Behavioral, Physiological and Anatomical Expressions of Age-Related Hearing Loss in the Laboratory Mouse: Original Papers Travaux originaux

Abstract
The auditory nerve isoelectric thresholds, in response to tone pips ranging from 5 to 20 kHz, are similar in the young C57BL/6 and CBA/J mice, although the latter genotype has somewhat more sensitive responses from 30 to 80 kHz. But their behavioral audiograms, obtained by the classical conditioning technique of Ehret, are very discrepant. Even though the behavioral audiogram of the CBA/J mouse can be predicted by measurements from its auditory nerve, the behavioral thresholds of the young C57BL/6 mouse are approximately 40 dB less sensitive than its electrophysiological measurements. Cochlear hair cell loss was not evident in young mice of either genotype. As the CBA/J approaches the end of its predicted life span, its auditory anatomy, physiology and behavior are not significantly altered; but those rare individuals who exceed their predicted life span by 40% finally develop hearing loss of a mixed nature. By contrast, the C57BL/6 mouse shows a relatively rapid decline of hearing as it ages. By 200 days of age, its auditory nerve responses are 30 dB less sensitive at 5 kHz, and 55 dB less sensitive at 30 kHz, than similar measures taken at adolescence. Over this same age span, its behavioral sensitivity has only declined by 15 and 25 dB at these two frequencies. Hair cell counts correlated poorly with both behavioral and electrophysiological auditory measures in the C57BL/6 mouse. Les seuils isoélectriques du nerf auditif, en réponse à des tone-pips de 1 ms pour des fréquences de 5 à 20 kHz, sont similaires chez les jeunes souris CBA/J et C57BL/6, bien que le premier génotype ait des réponses un peu plus sensibles entre 30 et 80 kHz. Mais leurs audiogrammes de comportement, obtenus par la méthode de conditionnement classique d'Ehret, sont très différents. Bien que l'audiogramme de comportement de la souris CBA/J puisse ětre prédit par des mesures de son nerf auditif, les seuils de comportement de la jeune souris C57BL/6 sont d'environ 40 dB moins sensibles que ses mesures électrophysiologiques. Aucune perte de cellules ciliées cochléaires ne fut constatée pour les deux génotypes. Quand la souris CBA/J approche de la fin normale de sa vie, son anatomie auditive, sa physiologie et son comportement ne sont pas altérés de manière significative. Mais les rares individus qui dépassent la longévité normale de 40% développent finalement des pertes d'audition d'une nature mixte. Par contre, la souris C57BL/6 montre un déclin relativement rapide de l'ouïe alors qu'elle vieillit. A l'aˇge de 200 jours, ses réponses du nerf auditif sont de 30 dB moins sensibles à 5 kHz, et de 55 dB moins sensibles à 30 kHz, que des mesures similaires prises durant l'adolescence. Durant cette měme période l'aˇge, la sensibilité de son comportement n'a decliné que de 15 et 25 dB à ces deux fréquences; on observe enfin une faible corrélation entre, d'une part, les mesures de comportement et les mesures électrophysiologiques, et, d'autre part, le nombre des cellules ciliées.Keywords
