Effects of amphotericin B and its methyl ester on plasma membranes ofCandida albicansand erythrocytes as examined by freeze-fracture electron microscopy

Abstract
Freeze-fracture electron microscopy of the plasma membranes of Candida albicans yeast cells and red blood cells treated with amphotericin methyl ester and amphotericin B showed that amphotericin B (50 μg ml-1) caused extreme aggregation of intramembranous particles on the protoplasmic fracture face of the C. albicans membrane, and a marked reduction of the density of intramembranous particles. On the other hand, the rearrangement of intramembranous particles induced by amphotericin methyl ester (50 μg ml-1) produced elevations of the particle-free membrane domains toward the outside of the cells, so that the particles were aggregated in linear furrows surrounding these elevations on the protoplasmic fracture face, and the corresponding ridges on the exoplasmic fracture face. The density of intramembranous particles was greatly reduced on the protoplasmic fracture face. Both polyenes produced only small changes in the erythrocyte membranes at the same concentration. These results suggest that amphotericin methyl ester affects the ergosterol-containing membranes more than amphotericin B, and that ergosterol has a higher sensitivity for these two polyene antibiotics than cholesterol. Le cryo-décapage des membranes plasmatiques de levures de Candida albicans et de globules rouges traités par l'amphotéricine méthyle ester ou par l'amphotéricine B, montre que l'amphotéricine B (50 μg ml-1) provoque une très forte agrégation des particules intramembranaires sur la surface protoplasmique fracturée de la membrane de C. albicans et une réduction marquée de la densité des particules intramembranaires. D'autre part, le réarrangement des particules intramembranaires, provoqué par l'amphotéricine méthyle ester (50 μg ml-1) entraîne une surélévation, vers l'extérieur des cellules, des régions de la membrane exemptes de particules, de telle sorte que les particules se trouvent agrégées dans des sillons linéaires autour de ces surélévations, sur la surface fracturée du protoplasme, ainsi que sur les arêtes correspondantes, sur la surface fracturée de l'exoplasme. La densité des particules intramembranaires est très réduite sur la surface fracturée protoplasmique. A la même concentration, les 2 polyènes ne produisent que de légers changements sur les membranes érythrocytaires. Ces résultats suggèrent que l'amphotéricine méthyle ester affecte plus les membranes contenant de l'ergostérol que ne le fait l'amphotéricine B et que l'ergostérol est plus sensible à ces deux antibiotiques que le cholestérol.This publication has 28 references indexed in Scilit:
