THE ROLE OF HUMAN COMPLEMENT IN THE LYMPHOCYTOTOXICITY TEST

Abstract
SUMMARY The apparently conflicting reports of the enhancing and inhibitory properties of fresh human serum in the cytotoxicity test are investigated and it is suggested that these anomalies are attributable to an imbalance between complement factors present in rabbit serum and fresh human serum. It is necessary to recognise the role played by human complement when screening for, standardising, and utilising cytotoxicity antisera for tissue typing, and its implication in crossmatching for transplantation. The use of a controlled amount of fresh human serum for a standardised cytotoxicity test is indicated and a mixture of rabbit serum and fresh human serum in proportions of 3:1, respectively, is suggested.
