Abstract
The tetrazolium technique has been used to study the mitochondrial and glycolytic enzymes in a skeletal muscle of the rabbit. The histological picture given by mitochondrial enzymes in C fibers appeared to show a diformazan distribution pattern similar to that of mitochondria demonstrated by other techniques. The pattern of diformazan distribution given by glycolytic enzymes was similar to the sarcoplasmic reticulum of Veratti. This was considered to be a false localization as it did not coincide with the cellular distribution of these enzymes as shown by differential centrifugation. The tetrazolium technique appears to have limited value in intracellular localization except for insoluble mitochondrial enzymes.
