Abstract
The amount of homologous Agrobacterium tumefaciens DNA carried in the genome of Scorzonera hispanica Crown-gall tissue cultures has been determined from the reassociation kinetics of 32P-labelled bacterial DNA in the presence of unlabelled Crown-gall DNA. Unrepeated DNA extracted from Crown-gall tissues was found to contain sequences homologous to the bacterial DNA; quantitative estimation shows the equivalent of about one Agrobacterium genome per copy of unrepeated Crown-gall DNA. It is concluded that an entire single genome of Agrobacterium is integrated into each haploid complement of a Crown-gall cell.Keywords
