Clinical Evaluation of Pagitane Hydrochloride in Parkinsonism

Abstract
The effect of Pagitane Hydrochloride in 23 cases of parkinsonism (due to previous encephalitis, 6 cases; arteriosclerosis, 4 cases; paralysis agitans, 9 cases; miscellaneous, 4 cases) was compared with that of Artane. The subjective and objective improvement was rated quantitatively and the side reactions were noted, it was concluded that the compound is an effective drug in reducing the rigidity and tremor. It can frequently be given in larger doses than similar compounds previously available, without producing unpleasant side reactions which would necessitate discontinuing medication. A tendency is noted for some of the patients to report greater improvement soon after beginning therapy. This is characteristic of parkinsonism and makes evaluation of the new remedy difficult. As has been noted previously by others, patients respond poorly to medical management if parkinsonism appeared late in life, tremor is the predominant symptom, and central nervous system changes attributable to arteriosclerosis are present. In these patients, mental confusion is frequently produced and the drug, if used at all, must be given with great caution.
