Inhibitory Action of Urate on p-Aminohippurate Secretion by the Isolated Rat Kidney

Abstract
We utilized an isolated perfusion rat kidney preparation to assess the influence of uric acid on p-amino-hippurate (PAH) secretion. In the absence of urate, PAHsecretion averaged 183 µg/min (0.94 µmol/min) at a perfusate PAHconcentration of 3.2 mg/dl (0.16 mmol/l), and was not influenced by increasing the perfusate PAHto 45 mg/dl (2.3 mmol/l). After addition of urate to the perfusate, PAHsecretion was 73% less at a perfusate PAHof 2.4 mg/dl (0.12 mmol/l), and changed very little following a 20-fold increase in perfusate PAH. The results raise the possibility that urate may depress renal organic anion secretion in vivo.
