Purification of Human Lymphotoxin

Abstract
This paper describes the first preparation of highly purified lymphotoxin, a soluble cytotoxin produced by lymphocytes. Lymphotoxin was produced in vitro by phytohemagglutinin-stimulation of human lymphocytes and was then purified greater than 2000-fold by a sequence of column chromatography (Sephadex G-150 followed by DEAE-cellulose) and preparatory acrylamide gel electrophoresis. Ten per cent of the cytotoxic activity was recovered in the final product. The results of the purification steps were evaluated by a new quantitative assay for cytotoxic activity.Keywords
