Pharmacokinetic studies of mezlocillin in newborn infants

Abstract
Sixty newborn infants with suspected or proven sepsis were treated with conventional doses of ampicillin and amikacin. A single im or iv dose of mezlocillin (75–100 mg) was substituted for a dose of ampicillin. Serum mezlocillin concentrations were determined at 0.5, 2, 4 and 8 h following the injection or infusion. The mean serum concentrations (mg/ml) were as follows: 0.5 h: 141.1; 2 h: 108.6; 4 h: 73.4; and 8 h: 32.9. The mean half life was 3.44 ( T½ ). Pharmacokinetic studies failed to demonstrate any significant differences between infants weighing ≤2000 g and those weighing >2000 g. Significant differences, however, were found in the T ½ of infants ≤ 7 days as compared to those T ½. Based on our data, suggestions for mezlocillin dosages in the newborn are made.Keywords
