Abstract
A technique for determination of the 25-OH-D content of serum directly without preparative chromatograpy has been developed and validated by comparison with reference assays of the 25-OH-D fraction separated by silicic acid chromatography. The direct assay technique selectively measures the 25-OH-D content of sample extracts even in the presence of a 3-5-fold excess of vitamin D as in the serum of hypervitaminotic D animals and subjects and without apparent interference from the dihydroxy metabolites of vitamin D. Meaningful clinical information can be obtained with this technique, allowing differentiation of vitamin D deficiency from other causes of hypocalcemia and hypervitaminosis D from other causes of hypercalcemia.Keywords
