A Slide Centrifugation Technic for Concentrating Blood Leukocytes and Nucleated Cells from Bone Marrow Blood

Abstract
A method for quickly obtaining leukocyte concentrates is proposed. The whole blood is hemolyzed by 1% ammonium oxalate and 0.2 ml. of hemolysate are centrifuged in a slide centrifugation apparatus (Cytocentrifuge) to obtain a monolayer of leukocytes in a small area. Hemolysis by saponin or cetrimide is not satisfactory as the cells are greatly damaged; centrifugation of plasma from sedimented blood is time consuming and leukocytes may be outnumbered by erythrocytes and platelets. Advantages of our technic are stressed, the main goals being the good preservation of the cells, the removal of undesired erythrocytes and platelets, and the rapidity of the procedures.Keywords
