Bestimmung stationärer Zwischenstoffkonzentrationen. I. Bestimmung stationärer DPN/DPNH-Konzentrationen in lebenden Zellen und Geweben

Abstract
Separate determinations were made of stationary DPN (diphosphopyridine nucleotide) and DPNH with alcohol dehydrogenase using the different stability of both compounds against acid and alkali for difference measurements at 340 m[mu]. Optimal conditions for deproteinization without alteration of the coenzyme were studied. The average DPN content of rat liver was found to be 317 [mu]g/g fresh tissue, that of DPNH 120 [mu]g/g; the ratio DPN/DPNH of 2.6/1 was altered, when quick deproteinization (15 seconds after death of rat) was replaced by freezing in liquid N. The DPN content of resting yeast cells was about 350 [mu]g/g fresh weight, the DPNH content was about 30 [mu]g/g. The DPN/DPNH ratio 12/1, fermentation and respiration increases the DPN and DPNH content up to 600 and 250 [mu]g/g.
