Abstract
Establishment of Lepidopteran cell lines and in vitro replication of insect‐pathogenic viruses. I. Mamestra brassicae cell lines and NPV‐replicationFive permanent cell lines have been isolated as monolayer cultures from organ explants and hemocytes of larvae. The morphological and proliferative characteristica of the lines were investigated. Sublines could be selected for permanent suspension culturing. In two cell lines replication of Autographa californica and M. brassicae NPV could be induced. Karyotype analysis of diploid and heteropolyploid cells have been performed. The aspects for virus production in vitro are discussed.Zusammenfassung: Aus Organexplantaten und Hämozyten‐Primärkulturen konnten fünf permanente Zelllinien von Mamestra brassicae isoliert werden. Das Material stammte aus L4/5‐Stadien. Morphologische Eigenschaften sowie Vermehrungsraten dienten als Parameter zur Charakterisierung der verschiedenen Zellinien. In zwei Zellinien konnte die Replikation von Autographa californica und M. brassicae NPV induziert werden. Karyotypanalysen wurden an diploiden und hetero‐polyploiden Zellen durchgeführt. Es konnten Verteilungskurven der Chromosomen pro Zelle erstellt werden. Aus den Monolayerkulturen gelang die Isolierung von permanenten Suspensionskulturen. Es werden Aspekte der Produktion von insektenpathogenen Viren in vitro diskutiert.Keywords
