Abstract
A rapid semimicro assay for lymphocyte activation in man is described. It is based upon the stimulation of protein synthesis (SPS) in peripheral blood lymphocytes cultured in serum-free medium with soluble antigens, phytomitogens or allogeneic lymphocytes. Cellular response to PHA is discernible in 4 hr and response to antigen in 16 to 24 hr. Stimulation in mixed lymphocyte cultures can be detected as early as 8 to 10 hr and invariably by 42 hr. The assay permits detection of lymphocyte activation earlier and with greater precision than the widely employed thymidine incorporation blast transformation system. Keywords
