THE ZYMOGRAM AS A TOOL FOR THE CHARACTERIZATION OF ENZYME SUBSTRATE SPECIFICITY

Abstract
The substrate specificity of 30 esterase and 2 phosphatase substrates using mouse liver as an enzyme source have been tested. The phosphatases in mouse liver, kidney, and ventral prostate have also been demonstrated. The small effect upon the zymogram obtained from the use of alpha and beta isomers of naphthyl acetate as substrates is pointed out and the equally small effect of the diazonium salt upon the zymogram obtained is reported. The differences in zymograms obtained by the use of the substrates are interpreted as due primarily to differing reaction rates on the part of the enzymes in the column rather than to all or none substrate specificity.
