THE GERMINATION OF MAIZE POLLEN

Abstract
A soln. containing 0.7% agar and 15% sucrose is held at 60[degree] C in a water bath and enough of this soln. transferred to a microscope slide to form a shallow droplet of approx. 1 cm. diam. The droplet is allowed to harden for 60 sec. at 20-25[degree] C and the pollen dusted on it from a knife blade held about 2 cm. above. The slide is immediately transferred to a chamber at 23[degree] C in which the rel. humidity is maintained at 90%. Germination counts can be made in 30 min. or the slide may be transferred to another moist chamber at 60[degree] C for later observation.
