INTRODUCTION OF A GENE FROM ESCHERICHIA COLI B INTO HFR AND F - STRAINS OF ESCHERICHIA COLI K-12

Abstract
The transfer of an auxotrophic allele (arg5-) from E. coli B to Hfr and F-strains of E. coli K-12 is described. The procedure involved conversion of the B mutant to an Flac+ genetic donor and mating of this male with an Hfr strain temporarily converted to the F- state. This method promises to be of general usefulness in transferring various types of genes from one strain to another and should facilitate the construction of multiple mutants having a set of desired marker genes. The transfer of the arg5-allele into an F-strain of K-12 has permitted mopping of this gene, which prior to this had not been possible because arg5-mutants had not been isolated in K-12. It was found that the presence of an Hfr gene excludes the effective transfer of the Flac+ episome during mating.Keywords
