Effect of Long Term Tolbutamide Treatment on Glucose Tolerance and Insulin Secretion in Maturity-Onset Diabetes Without Obesity

Abstract
Plasma glucose and immunoreactive insulin responses to oral glucose were measured before and after one week, two week and eight weeks of tolbutamide administration in ten nonobese maturity-onset diabetic patients. The glucose tolerance was significantly improved after the eight weeks treatment. The insulin response, very poor before treatment, was already significantly increased after one week and remained improved after eight weeks tolbutamide administration. It is concluded that glucose tolerance improvement after chronic tolbutamide therapy, in the defined category of patients, results from increased insulin response to glucose stimulus. 1 Work partially realized under Contract of the Ministère de la Politique Scientifique Belge within the framework of the Association Euratom, University of Brussels - University of Pisa.
