Abstract
Procedures were described for experiments utilizing a microculture system whereby it is possible to study the function of single helper T [thymus-derived] cells [mouse]. These are the elimination of the background anti-hapten response by preliminary anti-.theta. + guinea-pig complement treatment of the hapten-primed population; the removal of any contaminating hapten-specific B [bone marrow-derived] cells from the carrier-primed population by passage through nylon wool; irradiation of long-term primed nylon wool enriched helper cells to prevent cell division; the partition of individual helper cells by limiting dilution.
