Teflon-Sheathed, Fused Silica Connecting Tubing for Use in Microbore HPLC

Abstract
Teflon-sheathed fused silica (TFS) tubing is examined as a viable alternative to stainless steel (s.s) tubing in microbore HPLC. Compared to 1.007-in. i.d s.s. tubing, a 4% to 39% (depending on k′) increase in plate count is found using TFS tubing of 10 cm × 0.0025 in. i.d. Dispersion measurements with this tubing in a columnless system show a total (“system”) variance of 4.5 μl2, while the pressure drop is approximately 50 psi at 100 μlmin. It is shown that TFS tubing can be employed in a relatively long length (approximately 1 m) without introduced unacceptable extra-column disperation. Teflon-sheathed fused silica tubing is easy to handle and pressure stable up to 2500 psi.Keywords
