Effect of Interleukin-1 and 2 on Enzyme Secretion and Prostaglandin Fromation by Chondrocytes

Abstract
Following treatment of rabbit articular chondrocytcs with pure human IL-1 or conditioned media from macrophages a concentration-dependent massive increase of collagcnasc. neutral protcinasc and prostaglandin secretion, and a parallel decrease of plasminogen activator release can be observed. IL-2. indomcthacin or prostaglandin E2 agents known to interfere with the classical thymocytc proliferation assay, do not influence the chondrocytc response PMA reduces plasminogen activator and at 10 nM induces prostaglandin secretion from chondrocytcs but has no effect on neutral protcinasc release. Using a synthetic substrate for plasminogen activator and methylumbcllifcrylcdscin for neutral prolcmasc determination, the chondrocytc test system is simple, convenient, and could possibly be of value as a routine assay for measuring IL-1Keywords
