Physical Binding of Insulin by Gamma Globulins of Insulin-Resistant Subjects12

Abstract
Sera from insulin-resistant diabetic patients, but not of normal subjects or diabetic patients who were not insulin-resistant, demonstrated binding of added I131-labeled bovine insulin by gamma globulins when fractionated by a paper electrophoresis technique. The insulin binding capacity of this sera ranged from 0.05 to more than 20 mg/ml of serum and seemed to agree with other estimations of the degree of insulin resistance. Species specificity was demonstrated by the lack of interference with the binding when human insulin was added to the sera. It was felt that this binding represented the formation of an antigen-antibody complex in the sera of insulin-resistant subjects.
