Inhibition of Glucose and Tolbutamide-Induced Insulin Release by Iodoacetate and Antimycin A1

Abstract
Iodoacetate (5 HIM) inhibited both glucose-and tolbutamide-induced insulin release in the perfused rat pancreas and also isolated rat islets. In association with the iodaocetate inhibition of glucose-and tolbutamide-induced insulin release, islet ATP content decreased. Pyruvate, when added to either glucose or tolbutamide in the presence of iodoacetate, restored islet ATP content. Whereas with tolbutamide-induced insulin release pyruvate restored insulin secretion to normal in the presence of iodoacetate, no similar correction was observed with glucose. Pyruvate in the presence of glucose did not promote insulin release. Antimycin A inhibits both glucose-and tolbutamide-induced insulin release. (Endocrinology89: 169, 1971)Keywords
