Enzymatic Synthesis of 2′,3′-Dideoxyguanosine

Abstract
2′,3′-Dideoxyguanosine (ddG) can be conveniently synthesized in very high yields by utilizing the hydrolase activity of the ubiquitous mammalian enzyme, adenosine deaminase (ADA). The immediate precursor in this synthesis is 2-amino-6-chloro-2′,3′-dideoxypurine nucleoside, which is readily prepared from guanosine.
