Abstract
SUMMARY The conditions for the production of lymphotoxin from guinea pig lymph node cells stimulated with Phaseolus vulgaris phytohemagglutinin (PHA) are described. A comparison of various assay methods of lymphotoxin, including the incorporation of isotopically labeled compounds, standard cell counting, and the release of isotope from 51Cr-labeled cells, was made. The method utilizing 14C-amino acid or 3H-inositol uptake was the most satisfactory in both sensitivity and accuracy. The advantages and disadvantages of other assay methods are also described. Keywords
