Cold stage for electron probe microanalyser

Abstract
SUMMARY: Investigators wishing to examine and analyse biological specimens in the electron probe microanalyser (EPMA) in as near the original physiological and morphological state as possible have found that a cold environment is necessary to reduce the rate of sublimation and radiation damage to a low level.A low temperature cold stage has been designed and constructed for this purpose, to be used with a J.E.O.L. JXA 50A microanalyser. The cold stage will maintain the specimen holder at a temperature of 108 K for a period of 2 h before refilling of the liquid nitrogen Dewar flask becomes necessary. The fitting of a cold stage in place of the regular room temperature standard stage greatly increases the risk of contamination from the condensation of residual gases present in the specimen chamber. We have constructed an ami‐contamination plate (cold plate) that prevents this.Keywords
