Abstract
A tentative amino acid sequence of RNase U1 [EC 3.1.4.8] was derived by aligning tryptic and chymotryptic peptides obtained from the oxidized protein on the basis of maximal homology with the sequence of RNase T1 and with the aid of overlapping peptides. The sequence thus derived is different from that of RNase T1 in about 60% of the total residues. However, it was found to contain the amino acid sequences corresponding to those in RNase T1 involving His-40 and -92, Glu-58, and Arg-77 which have been implicated in the active site of RNase T1. These results strongly support the importance in the activity of these residues in RNase T1 and of the corresponding residues in RNase U1.Keywords
