Abstract
Requirements for maximum quantitative precision are developed for optical scanning of samples within TLC media. Baseline noise and variations in signal sensitivity are attributed to particle size distribution and layer thickness variations within the TLC media. Signal sensitivity variations are also traced to sample-to-sample differences in sample distribution within the TLC media. A table of densitometric and chromatographic requirements is given for maximum precision. These requirements are achieved in the Lightner Instrument Co. Automatic Thin Layer Analytical System (ATLAS). Precision data from ATLAS is presented which demonstrates the use of internal standards to obtain high precision.Keywords
