Analysis of Ascorbic acid in Human Plasma and Urine by HPLC With Electrochemical Detection

Abstract
The resolving power of high pressure liquid chromatography has been combined with the sensitivity of electrochemical oxidation to develop a method for determination of ascorbic acid in plasma and urine. A simple protein precipitation with 10% trichloroacetic acid and dilution in mobile phase (to which sodium metabisulfite has been added) preceded chromatograDhy. Sample collection and preparation procedures are designed to allow maximal recovery of ascorbic acid. As presented, a practical lower sensitivity of 1.0 ug/ml of plasma or urine with a total of 7 to 8 determinations per hour (unknowns and standards) is possible. The analysis of over 800 samples in a bioavailability study has demonstrated the reliability of the method.
