Abstract
Spinacin, which is isolated from Selachi liver, has the composition, C7H8O2N3 + 2 H2O. One hundred mg. of spinacin are mixed with 3 g. soda lime, and heated in a microcombustion tube in an atmosphere of H. The vapors are absorbed in dilute HC1 for 1 hr., and the soln. concentrated to dry ness in vacuo. The 4,5-methylimidazole is extracted from the residue with alcohol. The imidazole gives an intense Pauly diazo reaction and is precipitated as an Au salt (C4H6N2HAuCl4) with 30% chloroauric acid in conc. HC1. The Au salt is recrystallized from conc. HC1, dried and melts at 186[degree]-188[degree]. Since its m. p. is identical with that of the Au salt of synthetic 4.5-methylimidazole, the presence of an imidazole nucleus in spinacin is established. Spinacin is an imidazole derivative as are histidine, histamine, uro-canic acid, carnosine and ergothioneine. Whether or not spinacin has biological activity, has not been detd. because of the small amt. of spinacin available.
