Abstract
Tissue culture lines were established from tomato varieties susceptible and resistant to tomato race 0 of Phytophthora infestans. Inoculation of tissue culture pieces showed that at least partial resistance to the pathogen was maintained in the undiferentiated callus. An extraction procedure was developed for fractionation of fluorescent phenolic compounds. Chromatographic comparisons of phenolics from resistant and susceptible tissue culture lines, before and up to 6 days after inoculation, showed no significant differences in phenolic content. Ten days after inoculation there was an almost total loss of fluorescent phenolics from susceptible pieces. Comparisons of callus and leaf phenolics showed large qualitative differences between these two tissue types.
