Die papierchromatographische Trennung und Bestimmung der Uronsäuren

Abstract
A solvent is developed for separation of glucuronic, mannuronic and galacturonic acids, in which the spots are discrete without tail formation or excessive spreading. The composition is pyridine-ethyl acetate-acetic acid - water in the ratio 5:5:1:3, in a saturated atmosphere arising from a mixture of pyridine-ethyl acetate - water (11:40:6). Rf values for a number of substances including keto-glucuronic acids and glucosamine are given. Separation of mannurone, glucurone and gulurone can be achieved in pyridine-ethyl acetate-water (11:40:6). The stability towards acid hydrolysis of the uronic acids was investigated; there is appreciable loss even after 5 hours heating at 100[degree]C with 0.1 [image] HC1. In the cource of hydrolysis of uronic acid containing natural products diuronic acids may be formed which are largely resistant to further hydrolysis. The chromatography of some of these was also investigated.
