Abstract
A solvent is developed for separation of glucuronic, mannuronic and galacturonic acids, in which the spots are discrete without tail formation or excessive spreading. The composition is pyridine-ethyl acetate-acetic acid - water in the ratio 5:5:1:3, in a saturated atmosphere arising from a mixture of pyridine-ethyl acetate - water (11:40:6). Rf values for a number of substances including keto-glucuronic acids and glucosamine are given. Separation of mannurone, glucurone and gulurone can be achieved in pyridine-ethyl acetate-water (11:40:6). The stability towards acid hydrolysis of the uronic acids was investigated; there is appreciable loss even after 5 hours heating at 100[degree]C with 0.1 [image] HC1. In the cource of hydrolysis of uronic acid containing natural products diuronic acids may be formed which are largely resistant to further hydrolysis.
