A SIMPLE METHOD FOR PREPARATION AND STAINING OF THE WHOLE PACINIAN CORPUSCLE

Abstract
A simple inexpensive procedure is described for the preparation of whole mounts of Pacinian Corpuscles from the cat mesentery. The whole mesentery from an anaesthetized cat is removed and kept in a jar containing 1% silver nitrate in distilled water for 4 to 6 hrs. Then tissue is removed, washed in 6 changes of distilled water, each change lasting 10 min., and the Pacinian Corpuscle is isolated from the mesentery under the dissection microscope. The nuclei can be counterstained by using 1% aqueous cresyl violet, dehydrating in ascending grades of alcohol and mounting in D. P.X. Series of superficial lamellae can be removed to demonstrate that deeper lamellae are also made up of continuous layers of flat squamous cells. The silver nitrate stains the cellular borders and the cresyl violet the nucleus.
