The effect of added phosphate compounds in concentrations from 1 to 3 mM was studied in human erythrolysate. 2.3-diphosphoglyceric acid (2.3-DPG) and ATP decreased the oxygen affinity of hemoglobin as indicated by an increase in the T50 value. The effect of 2.3-DPG on a molar basis is more pronounced than that of ATP. AMP, ADP, NADH and NADPH had essentially no effect on the oxyhemoglobin dissociation curve (ODC) in the erythrolysate. The preliminary results reported here support the concept that variations in the concentration of 2.3-DPG and ATP in the erythrocyte within the physiological range could play a role for the oxygen affinity of hemoglobin in vivo.Keywords
